The remake is being written by Brooklyn Nine-Nine alum Ayesha Carr, which marks her feature film debut.
The plot will be a modern setting of the original, which revolves around two strangers, who must team up to tackle the gantlet of holiday travel disasters and make it home to their loved ones.
The original "Planes, Trains, and Automobiles" starred Steve Martin and John Candy.
Hart confirmed the news on Instagram, saying: "I've been patiently waiting for the right project to team up with my brother with," he wrote. "We found it and are developing it together with our teams. This will be huge for us and the city of brotherly love aka Philadelphia..... let's goooooo."
