Djokovic, who is chasing his first Olympic gold medal, will take the Australia’s Matthew Ebden in the opening round.
On the other hand, Nadal, who won the gold medal in Beijing in 2008,will face Hungary’s Marton Fucsovics.
Roland Garros and Wimbledon Champion Carlos Alcaraz will open his campaign against Lebanon’s Hady Habib.
The German Alexander Zverev, the holder of a gold medal, will face Spain’s Jaume Munar in the first round.
Hubert Hurkacz and World No. 16 Holger Rune withdrew from the tournament.
Andy Murray, the only man to win two Olympic gold medals in singles, withdrew from the singles event minutes before the draw ceremony.
He will only play doubles in Paris, teaming up with Dan Evans. The duo will face Japan’s Taro Daniel and Kei Nishikori in the first round.
For women, Iga Swiatek will face Irina-Camelia Begu in the opening round of the women’s singles, while world number two Coco Gauff will take on Ajla Tomljanović.
Naomi Osaka will face Angelique Kerber in the women's singles.
